Silk fibroin has good properties such as flexibility, sustained release, air permeability, moisture permeability and tensile strength. After processing, it can be obtained in different forms such as fiber, solution, powder, film, and gel.

    Silk fibroin is a kind of powdery polymer silk fibroin protein obtained by selecting the silk fibroin after high pressure degumming, processing it by a special process, and then using physical methods to pulverize it. The molecular weight is approximately 250,000. Tasteless, odorless, slightly soluble in water, insoluble in alcohol ether.

These favorable properties are due to its unique physicochemical properties.

     

    Chemical composition and structure of silk fibroin

     

    The relative molecular weight of silk fibroin is relatively large, and the molecular structure and intermolecular interactions are very complex. Generally speaking, the silk fibroin molecular chain consists of three subunits, namely the heavy chain (H-chain), the light chain (L-chain) and the glycoprotein. P25 composition. The heavy and light chains are linked by disulfide bonds, and then bind to glycoprotein P25 by non-covalent interactions such as hydrophobic bonds.

    The heavy chain (H chain) consists of 5236 amino acid residues with a molecular weight of 391kDa; the light chain (L chain) consists of 266 amino acid residues with a molecular weight of 28kDa: the P25 protein is similar in size to the light chain, about 25kDa. The ratio of heavy chain, light chain and P25 in silk fibroin is about 6:6:1.

    Silk fibroin of Bombyx mori is composed of 18 amino acids, of which glycine (Gly, 43%), alanine (Ala, 30%) and serine (Ser, 12%) account for about 85%.

    The main crystal structures of silk fibroin are Silk I and Silk II. A small and unstable Silk III structure was also present in the regenerated silk fibroin solution at the air/water interface. Silk I includes helical and other non-β-sheet conformations, while Silk II structures mainly refer to antiparallel β-sheet conformations.
