Confocal is an advanced fluorescence microscope. Its advanced features are:
① A photomultiplier tube (PMT) is used to collect signals at a single point, and this point is scanned to form an area imaging; a fluorescence microscope uses a camera to directly perform area imaging
②Confocal has a “pinhole” in front of the detector, so signals above and below the focus can be eliminated, achieving an image effect similar to “CT”
③Confocal can excite samples with multiple lasers at the same time, and dyes of different colors correspond to different detectors, thereby achieving "1 corresponding laser + 1 corresponding detector" to form "1 channel", and multiple channels realize multi-color Fluorescence signal collection;
④ Because fast laser switching and corresponding detector switching can be performed for a single point, confocal can achieve "sequential" scanning at a microscopic level. In addition, the laser has good monochromatic type, thus achieving rapid multi-color scanning. Avoid color bleed while shooting
⑤Fluorescence microscope has low switching speed due to the color filter block. The filter wavelength installed on it cannot be perfect, so the multi-color speed is low and color cross-over is easy.
