Iron Dextran Solution Preparation

Ⅰ.Determination the Fe content of the iron dextran powder. (obtain the content from the COA reported by Taoyuan Veterinary Drugs Factory).

Ⅱ.Calculate the amount of water consumption for dissolved & preservative (phenol)

A:  water consumption for dissolved  in  KG

B:  weight of iron dextran powder    in  KG

C:  content of iron                 in  % (obtain from the COA)

D:  Phenol consumption            in  KG

1. Specification 5% ( content of Fe: 50mg/ml±5%)

Calculation Formulation for specification 5% :A=B*C*21.4 – B

D= [(B + A)]*0.05%

For example, make 50kg iron dextran powder into 5% solution, the water consumption for dissolved is  50kg*44.8%*21.4-50kg=429.36 kg 

429.36kg is the total water add into the iron dextran powder.

2. Specification 10% ( content of Fe: 100mg/ml±5%)

Calculation Formulation for specification 10% :A=B*C*11.5– B

D= [(B + A)]*0.05%

3. Specification 20% ( content of Fe: 200mg/ml±5%)

Calculation Formulation for specification 20% :A=B*C*6.45– B

D= [(B + A)]*0.05%

Ⅲ.According the calculation result, soak the powder in 50% of the dissolved water(which must be water for injection) for 48 hours. Put the 40% of the dissolved water into enamel reactor （or acid & alkali resistance container）, stir, raise the temperature to 90 ~ 95 ℃. Stir the soaked iron dextran and join it into the enamel reactor(or acid & alkali resistance container）.Cleaning the soaking container with the rest of the amount water, the washed water join into the enamel reactor(or acid & alkali resistance container）, Insulation around 95 ℃ and stir until dissolved completely (it will take about 3 ~ 4 hours), make sure it dissolved completely, then cooling to 40 ℃,add quantitative phenol, stir completely, cooling below 30 ℃, Sampling and determination, check all technical indicators are compliance, discharge the iron dextran solution from the container. Filter the impurity, determination the technical indicators again, compliance can be repackaging.
