    About The Detailed Introduction Of Glucose Oxidase?

     

    Glucose oxidase powder (GOD) is obtained by deep ventilation fermentation with Penicillium aureus, precipitating it with ethanol and acetone and adsorbing it with kaolin or aluminum hydroxide, and then salting out and refining it with ammonium sulfate. It can also be produced from Penicillium spp. and Aspergillus niger. Almost white to light yellow powder, or yellow to brown liquid. Soluble in water, the aqueous solution is generally light yellow. Almost insoluble in ethanol, chloroform and ether.
    It is an important industrial enzyme in the food industry. It is widely used in deoxidation of wine, beer, juice, milk powder and other foods, flour improvement, and prevention of food browning. It is also widely used in rapid food detection and biosensors. GOD is widely distributed in animals, plants and microorganisms. Microorganisms grow and reproduce quickly and have a wide range of sources. They are the main source of GOD production. The main production strains are Aspergillus niger and Penicillium.
    Glucose oxidase is an enzyme found in molds such as Penicillium notatum and in honey. It can catalyze the reaction of D-glucose + O2D-gluconic acid (delta-lactone) + H2O2. EC1.1.3.4. The enzyme of Penicillium specificum (P. notatum) has attracted attention due to its obvious antibacterial properties. Therefore, it is also called glucose oxidase (notatin). It is now clear that the antibacterial property is due to the sterilizing properties of H2O2 generated by the reaction. The purified product contains 2 molecules of FAD. As an electron acceptor, in addition to O2, it can also react with 2,6,dichlorophenol and indophenol. This enzyme is specific for glucose.

     

    Enzyme Structure


    GOD is a homodimer molecule containing 2 flavin adenine dinucleotide (FAD) binding sites. Each monomer contains 2 completely different regions: one is non-covalently but tightly bound to part of FAD, mainly β-sheet; the other is bound to the substrate β-D-glucose, supported by 4 α-helices and 1 anti- Parallel beta sheets.

     

    Enzymatic Properties

     


    High-purity GOD is a light yellow crystal that is easily soluble in water and can be made into liquid preparations. GOD is completely insoluble in organic solvents such as ether, chloroform, butanol, pyridine, glycerin, and ethylene glycol. The relative molecular mass of GOD is generally around 1.5×105, and each molecule of enzyme contains 2 molecules of FAD. The pH range is 3.5 to 6.5, and the optimal pH is around 5.0. It will be rapidly deactivated at pH >8.0 or pH <3.0 in the absence of a protective agent. The operating temperature range of GOD is generally 30 ~ 60 ℃. Solid enzyme preparations can be stored at 0°C for at least 2 years and at -15°C for 8 years.
