Clozotinib is an oral aminopyridine inhibitor that is an inhibitor of receptor tyrosine kinase (ALK) and c-Met/hepatocyte growth factor receptor (HGFR), exhibiting anti-tumor activity. Clozotinib competitively binds to ALK kinase and ALK fusion protein in an ATP competitive manner and inhibits them. In addition, clotozantinib can also inhibit c-Met kinase and disrupt the c-Met signaling pathway. Overall, this medication can inhibit the growth of tumor cells. ALK belongs to the insulin receptor superfamily and plays an important role in the development of the nervous system. ALK dysregulation and gene rearrangement are associated with a range of tumors.
