Active targeting is achieved through receptor-specific interaction with the target cell and is dependent on the targeting ligand. These ligands include small molecule ligands, carbohydrates, peptides, antibodies, or aptamers. Depending on the physicochemical properties of these ligands, two methods can be used to bind them to lipid nanoparticles:
"One-step" assembly: All structural lipids, auxiliary lipids and targeting ligands are assembled into lipid nanoparticles in one go. This approach introduces ligands at the same time as lipid nanoparticles to achieve targeted action (Figure a).
"Post-insert" preformed nanoparticles: First, the preformed ordinary lipid nanoparticles are prepared. The targeting ligands are then inserted into these nanoparticles. This method achieves targeting by introducing ligands into the nanoparticles so that the nanoparticles that have already been formed have targeting capabilities (Figure b).
