Concept and classification of antimicrobial peptides
Antimicrobial peptides are short peptides with antibacterial activity. In 1975, a bactericidal peptide was obtained from the pupae of H. occidentalis by G. Boman of Germany and named cecropin. Since then, many antimicrobial peptides have been isolated and purified. The amino acid primary structures and gene sequences of some antimicrobial peptides were identified. In the 1980s, the research of antimicrobial peptides mainly focused on large insects. Since 1990s, in addition to the research on large insects, it has been extended to some small and medium-sized insects and other invertebrates and vertebrates. Antimicrobial peptides have become a hot spot in immunology and biology. So far, many proteins and polypeptides with antibacterial effect have been found in different animal tissues. The structures of more than 70 kinds of antimicrobial peptides have been determined, and the concept of antimicrobial peptides has been greatly expanded.
According to the structure of antimicrobial peptides, they can be divided into five categories
(1) Single chain α - helix without cysteine residues or peptides composed of several α - helices connected by random coils;
(2) Antimicrobial peptides rich in some amino acid residues but not cysteine residues;
(3) An antibacterial polypeptide containing a disulfide bond;
(4) Antimicrobial peptides with two or more disulfide bonds and β - fold structure;
(5) Antimicrobial peptides derived from other large peptides with known functions.
Among them, cecropins and magainins isolated from Xenopus laevis belong to the first class of antimicrobial peptides, which are also known as cecropin antimicrobial peptides. At present, the research on these antimicrobial peptides is more in-depth.
