General peptide sequences are written in two forms: one-letter and three-letter forms, either of which the peptide structure expressed is exclusive in order to prevent divergence. At present, the international unified standard is written that the N-terminal (nitrogen-terminal) of the peptide is on the leftmost side and the C-terminal (carbon-terminal) of the peptide is on the rightmost side.
Single-letter Writing Form of Peptides
What is a single letter?
Simply speaking, a letter represents an amino acid, which is generally indicated in capital letters, such as A for alanine and C for cysteine. If it is a lowercase letter, the corresponding amino acid is D-type, such as the lowercase letter a represents D-type alanine, and the lowercase letter C represents D-type cysteine.
EXAMPLES OF PEPTIDE SINGLE-LETTER Writing
1. Peptide 1 sequence: AGYFKVCM, a total of eight amino acids, the letters are capitalized in English, indicating that all the amino acids are purely natural L-type amino acids.
2. Peptide 1 sequence: AGYFKVCM can also write H-AGYFKVCM-OH, H-on the left represents the N-terminal amino group, which can be written or omitted. The -OH on the right represents the carboxyl group at the C-terminus, which can either be written or omitted. But generally for the sake of beauty and neatness, H- and -OH are written or not.
3. Ppeptide 2 sequence: AGYFkvcm, a total of eight amino acids, the four amino acids on the right (C-terminal) are lowercase letters, indicating that the corresponding four amino acids are D-type amino acids with L-type symmetry.
4. Peptide 4 sequence: AGYFKVCM-NH2, different from peptide 1, there is much -NH2 on the right side, which represents the C-terminal amidation.
Three-letter Writing Form of Peptides
What are three letters?
That is, three letters represent an amino acid, and the three letters are one-to-one correspondence with the letters mentioned above.Of the three letters, the first letter is capitalized, the second and third letters are lowercase, such as Ala also represents alanine, Cys represents cysteine.
Examples of Three-Letter Writing of Peptides
1. The three letters corresponding to the peptide 1 sequence are written as: H-Ala-Gly-Tyr-Phe-Lys-Val-Cys-Met-OH.
2. The corresponding three letters of peptide 3 sequence are written as: H-Ala-Gly-Tyr-Phe-Lys-Val-Cys-Met-NH2, C-terminal amidation.
Writing of unique amino acids. Some common writings are introduced below.
1. The writing of N-terminal end, generally acetic acid end, is written as Ac-Ala-Gly-Tyr-Phe-Lys-Val-Cys-Met-OH.
2. Writing of phosphorylated amino acids:
H-Ala-Gly-pTyr-Phe-Lys-Val-Cys-Met-OH
H-Ala-Gly-Tyr (PO3H2)-Phe-Lys-Val-Cys-Met-OH, numbers 3 and 2 can be subscripted.
