2. Preparation of Cationic Liposome Microbubble Ultrasound Contrast Agent and Preliminary Study on Gene Transfection in Vitro

A cationic liposome microbubble ultrasound contrast agent that can be observed in real time by conventional ultrasound was prepared to provide a carrier that can be observed in real time, thus providing certain technical support for gene carrying microbubble combined with ultrasound targeted therapy.
Three lipids, DPPC,1, 2-diolenoxy-3-trimethylaminopropane (DOTAP) and distearoylphosphatidylethanolamine-polyethylene glycol 2000(DSPE-PEG2000), were mixed in a certain proportion to prepare liposome microbubble ultrasonic contrast agent by thin film dispersion method. The morphology of microbubbles was observed under a light microscope, the particle size distribution of microbubbles was detected by a laser particle size analyzer, and the development effect of microbubbles in vivo and in vitro was observed by a medical ultrasonic diagnostic instrument. In vivo development effect was compared with commercial ultrasound contrast agent (sonovine) to observe the difference. The ability of liposome microbubbles to carry PGL3-control gene was detected by DNA gel electrophoresis, and the transfection effect of the microbubbles in vitro was analyzed.
Results: The average particle size of liposome microbubbles prepared by mixing DPPC,DOTAP and DSPE-PEG2000 was 6.969μm. In vitro and in vivo development effect of ultrasound contrast agent was good, and there was no statistical difference in the development duration of large arteries in vivo compared with sonovi (P >0.05). DNA gel electrophoresis experiments showed that the self-made cationic liposome microbubble ultrasound contrast agent had a better ability to carry PGL3-control gene, and the carrying ability was significantly better than that of Sonovi. Meanwhile, sonovi was also found to have a very weak ability to carry PGL3-control gene. In vitro cell experiments confirmed that the self-made liposome microbubbles significantly improved the transfection efficiency of PGL3-control in COS-7 cells (P <0.01).
DPPG is used as injection grade dipalmitoylphosphatidylglycerol sodium, a fully synthetic phospholipid, which needs to be shaded, sealed and refrigerated to avoid co-storage with strong oxidizing substances.
