1. Triocylglyceride for Depocyt®
Cytarabine liposome, developed by Skypharm Inc., was marketed in the United States in 1999 for the treatment of Liu meningitis. Incorporating cytarabine packs into liposomes reduced the number of dressings from once every two days to once every two weeks, and significantly improved patient compliance. The phospholipids of Chu formula were composed of DOPC, DPPG, cholesterol and glycerol trioleate. The product was a 5mL saline suspension, each containing 50mg cytarabine, 28.5mGDOPC, 5mGDPPG, and 22mg cholesterol.
2. Triocylglyceride for use in DEPODUR ®
Ka Sulphate liposomes, developed by Skypharm Inc., were approved by the FDA in 2004 for preoperative anesthesia or intraoperative analgesia. The phospholipids of Chu formula were composed of DOPC, DPPG, cholesterol, triocylglyceride and trioleate. Each product contains 10mg/mL Ka sulfate, 4.2mg/mL DOPC, 0.9mg/mL DPPG, 0.3mg/mL Trioctanate, 3.3mg/mL Cholesterol and 0.1mg/mL Trioleate.
3. Triocylglyceride is applied to EXParel ®
Pacira Pharmaceuticals, Inc. 's bupivacaine liposome was approved by the U.S. Food and Drug Administration in 2011 for use in postoperative analgesia at surgical sites. The liposome extends drug release up to 72 hours. Chu uses DEPC, a new PC-based phospholipid excipient, which is the only commercially available product based on DepofoamTM technology. The phospholipids of Chu formula consist of DEPC, DPPG, cholesterol and trioctylyceride. Each bottle contained bupivacaine 13.3mg/mL, DEPC 8.2mg/mL, DPPG 0.9mg/mL, cholesterol 4.7mg/mL, and trioctylyceride 2.0mg/mL.
